STEELE, R., B. WINKLER, AND N. ALTSZULER. Inhibition by infused glycerol of gluconeogenesis from other precursors. Am, J. Physiol. 22 l(3) : 883-888. 1971 .-Plasma glycerol contributes 20% of the carbon of glucose released to the blood by the liver in the 6to 7-dayfasted dog (shown by use of glycerol-UJ4C). This increases to 63y0 when bulk glycerol infusion increases total glycerol uptake from plasma threefold. In the postabsorptive state, plasma glycerol contributes 7y0 of the carbon of released glucose; this increases to 55y0 when bulk glycerol infusion increases total glycerol uptake eightfold. Total glucose release (shown by use of glucoseJ4C) increases only at the highest rates of glycerol infusion and then by only 207& It is demonstrated that the contribution of other-than-plasmaglycerol carbon to the immediate precursors of the glucose released to the blood must decrease when the bulk glycerol is infused in order for the fractional contribution of plasma glycerol carbon to the released glucose carbon to rise to the observed levels. In the long-fasted dog, where hepatic glycogen is not a glucose source, the contribution of carbon to the glucose precursors from gluconeogenie sources other than glycerol must decrease when bulk glycerol is infused.

Phenethylbiguanide and the inhibition of hepatic gluconeogenesis in the guinea pig.
1. Phenethylbiguanide inhibits the synthesis of phosphoenolpyruvate from malate or 2-oxoglutarate by isolated guinea-pig liver mitochondria. This inhibition is time- and concentration-dependent, with the maximum decrease in the rate of phosphoenolpyruvate synthesis (80%) evident after 10min of incubation with 1mm-phenethylbiguanide. 2. The phosphorylation of ADP by these mitochondria is also in...
متن کاملThe inhibition of gluconeogenesis following alcohol in humans.
Accurate quantification of gluconeogenic flux following alcohol ingestion in overnight-fasted humans has yet to be reported. [2-13C1]glycerol, [U-13C6]glucose, [1-2H1]galactose, and acetaminophen were infused in normal men before and after the consumption of 48 g alcohol or a placebo to quantify gluconeogenesis, glycogenolysis, hepatic glucose production, and intrahepatic gluconeogenic precurso...
متن کاملRenal substrate exchange and gluconeogenesis in normal postabsorptive humans.
Release of glucose by the kidney in postabsorptive normal humans is generally regarded as being wholly due to gluconeogenesis. Although lactate is the most important systemic gluconeogenic precursor and there is appreciable net renal lactate uptake, renal lactate gluconeogenesis has not yet been investigated. The present studies were therefore undertaken to quantitate the contribution of lactat...
متن کاملRole of fructose 2,6-bisphosphate in the control by glucagon of gluconeogenesis from various precursors in isolated rat hepatocytes.
Hepatocytes from overnight-starved rats were incubated with 1-20 mM-fructose, -dihydroxyacetone, -glycerol, -alanine or -lactate and -pyruvate with or without 0.1 microM-glucagon. The production of glucose and lactate was measured, as was the content of fructose 2,6-bisphosphate. The concentrations of fructose (below 5 mM) and dihydroxyacetone (above 1 mM) that gave rise to an increase in fruct...
متن کاملEffect of insulin on the utilization of propionate in gluconeogenesis in sheep.
The effects of insulin on the utilization of propionate in glucose synthesis were studied in fed and fasted sheep. Insulin was infused at 0.40 microU/h into the mesenteric vein. Glucose was infused to prevent hypoglycaemia. The rate of incorporation of [2-14C]propionate into glucose was determined before and during insulin infusion. After 150 min of insulin infusion endogenous glucose synthesis...
